Removing of the N-terminal methionine improves the sweetness of the recombinant expressed sweet-tasting protein brazzein and its mutants in Escherichia coli
The sweet-tasting protein brazzein is the smallest candy protein with excessive candy efficiency. Overexpression of this protein in a heterogenous host is a vital means for its manufacturing in meals trade. On this research, the gene of minor kind brazzein was cloned into the pET-SUMO vector with optimized codon utilization and expressed in E. coli BL21-CodonPlus (DE3)-RIL.
The recombinant protein in absence of the N-terminal methionine displayed a sweetness threshold about 1.5 μg/ml, which is the sweetest brazzein protein reported to date. The surprising candy efficiency of the protein was validated by a sequence of mutants (7Val → Arg, 9Glu → Lys and 9Glu → Asp), wherein E9K exhibited about 50% enhancement of sweetness than the wild sort. The superior sweetness of recombinant brazzein and its sweeter mutants counsel their potential purposes in meals and drinks.
PRACTICAL APPLICATIONS: The sweet-tasting proteins are pure, low-, or non-caloric and nutritive, exhibiting to be promising replacers of sugars and synthetic sweeteners, and can be utilized as candy components within the fields of meals, drugs, and biotechnology. Within the current research, we report that the recombinant brazzein protein expressed in E. coli reveals superior and improved sweetness than these beforehand reported.
Moreover, sweeter mutants had been obtained with the expression process. The superior sweetness of recombinant brazzein and its sweeter mutants counsel their potential purposes in meals, drinks, and different fields.

TM Yeast Pst1 Recombinant Protein |
|||
95-151 | ProSci | 0.05 mg | EUR 322.25 |
Description: Protoplast secreted protein 1 (Pst1) is an anchored plasma membrane protein. Pst1 was previously identified as a protein secreted by yeast regenerating protoplasts, which suggests a role in cell wall construction (1). Pst1 is a protein with 444 amino acids and is attached to yeast cell wall via a glycosylphosphatidylinisotol (GPI) anchor (2). Pst1 contains 15 potential N-linked glycosylation sites and is heavily glycosylated. It migrates at around 200 kDa on SDS-PAGE when produced in wild type S. cerevisiae. Our Pst1 protein was expressed in a genetically manipulated triple-mutant (TM) S. cerevisiae stain (Δoch1 Δmnn1 Δmnn4), which results in the production of sole Man8GlcNAc2 carbohydrate structures and Pst1 migration at approximately 100 kDa. When produced in the TM yeast, Pst1 can be recognized by several HIV-1 broadly neutralizing antibodies, including 2G12 and recently identified glycan-specific PGT antibodies (3). Among several heavily N-glycosylated TM yeast glycoproteins, Pst1 shows high affinity for 2G12 and efficiently inhibits gp120 interactions with 2G12 and DC-SIGN, and it also blocks 2G12-mediated neutralization of HIV-1 pseudoviruses (4). |

Description: Neuronal protein that plays several roles in synaptic activity such as regulation of synaptic vesicle trafficking and subsequent neurotransmitter release. Participates as a monomer in synaptic vesicle exocytosis by enhancing vesicle priming, fusion and dilation of exocytotic fusion pores. Mechanistically, acts by increasing local Ca2+ release from microdomains which is essential for the enhancement of ATP-induced exocytosis. Acts also as a molecular chaperone in its multimeric membrane-bound state, assisting in the folding of synaptic fusion components called SNAREs (Soluble NSF Attachment Protein REceptors) at presynaptic plasma membrane in conjunction with cysteine string protein-alpha/DNAJC5. This chaperone activity is important to sustain normal SNARE-complex assembly during aging. Plays also a role in the regulation of the dopamine neurotransmission by associating with the dopamine transporter (DAT1) and thereby modulating its activity. |

Description: Alpha-synuclein (Snca) belongs to a family of proteins including a-, b-, and g-synucleins. Alpha-synuclein has been found to be implicated in the pathophysiology of many neurodegenerative diseases, including Parkinson's disease (PD) and Alzheimer's disease. Manyneurodegenerative diseases has shown that alpha-synuclein accumulates in dystrophic neurites and in Lewy bodies. The function of alpha-synuclein is closely correlated with its three-dimensional structure, especially for proteins important in the pathogenesis of neurodegenerative diseases. Alpha-synuclein is a dynamic molecule whose secondary structure depends on the environment. For example, it has an unfolded random coil structure in aqueous solution, forms a-helical structure upon binding to acidic phospholipid vesicles, and forms insoluble fibrils with a high b-sheet content that resemble the filaments found in Lewy bodies. Also, alpha-synuclein was known to associate with 14-3-3 proteins including protein kinase C, BAD, and extracellular regulated kinase, and overexpression of alpha-synuclein could contribute to cell death in neurodegenerative diseases. |
